GLC determination of quinaldine residue in fish

A procedure for the determination of quinaldine residue in various fish tissues is described. Homogenized tissues are extracted with hexane-ethyl ether, the extracts are concentrated by partitioning through 0.1N sulfuric acid, and the residues are measured by alkali flame ionization gas chromatography. Muscle tissues containing from 0.01 to 10.0 ppm quinaldine were successfully analyzed with recoveries from 75 to 100%.

UV identification and quantitative measurement of quinaldine residues in fish

A method for the confirmatory analysis of quinaldine residue in fish is described. The method utilizes the same extraction and cleanup procedure as the GLC method reported previously. The lower limit of sensitivity for quinaldine residue in fish muscle is 0.01 ppm. Identification is accomplished by comparing the UV spectra of the sample and a standard solution of quinaldine.

Residues of isobornyl thiocyanoacetate (Thanite) and a metabolite in fish and treated ponds

Isobornyl thiocyanoacetate (Thanite) is an insecticide that induces a surfacing response in fish and therefore has been considered to have potential as a fish collection agent. Analyses for residues of Thanite in carp (Cyprinus carpio) and largemouth bass (Micropterus salmoides) exposed to chemical yielded only a trace of the parent compound. A metabolite, isobornyl a-(methylthio)acetate, was isolated and indentified by GC-MS,and a reference standard for the metabolite was synthesized. Residues of the metabolite were present in largemouth bass muscle tissue within 1 h after exposure to Thanite. The metabolite was also observed in the muscle, blood plasma, and bile of carp. Residues of the metaboliteare rapidly elimanated after the fish are transferred to Thanite-free water. Residues of Thanite in water, algae, and from soil from ponds treated with Thanite declined to undetectable levels within 28 days after treatment.

The influence of pH on the efficacy and residues of quinaldine

Quinaldine, an anesthetic for fish, loses its effectiveness in solutions having pH values less than 6. Measured quantities of un‐ionized quinaldine in solution compared favorably with calculated values at selected pHˈs. Quinaldine residues in largemouth bass (Micropterus salmoides) anesthetized at various pHˈs verify that only the un‐ionized portion in solution enters the fish. Quinaldine residues in fish and un‐ionized quinaldine in solution were measured by gas chromatography.

Three thousand eyed coho salmon (Oncorhynchus kisutch) eggs from Lake Michigan stock were sent by the Department of Natural Resources to the Fish Control Laboratory, La Crosse, Wis., on January 15, 1969, for use in evaluating candidate fish-cnotrol chemicals.
